Synthesizing the recombinant Human GABARAPL1 protein generally involves integrating the DNA fragment that encodes the Human GABARAPL1 protein (1-117aa) into a plasmid, introducing the recombinant plasmid into e.coli cells, followed by the selection and culturing of positive e.coli cells, induction of protein expression, and subsequent cell lysis. A N-terminal GST tag is fused to the protein. The protein is purified through affinity purification, and SDS-PAGE analysis is conducted to confirm the presence of the protein and determine its purity. The protein's purity surpasses 90%.

Target Name
GABARAPL1
Target Full Name
Gamma-aminobutyric acid receptor-associated protein-like 1
Target Function
Ubiquitin-like modifier that increases cell-surface expression of kappa-type opioid receptor through facilitating anterograde intracellular trafficking of the receptor. Involved in formation of autophagosomal vacuoles. While LC3s are involved in elongation of the phagophore membrane, the GABARAP/GATE-16 subfamily is essential for a later stage in autophagosome maturation. Through its interaction with the reticulophagy receptor TEX264, participates in the remodeling of subdomains of the endoplasmic reticulum into autophagosomes upon nutrient stress, which then fuse with lysosomes for endoplasmic reticulum turnover.
Target Subcellular Location
Cytoplasmic vesicle, autophagosome. Cytoplasmic vesicle membrane; Lipid-anchor. Cytoplasm, cytoskeleton. Endoplasmic reticulum. Golgi apparatus.
Target Protein Families
ATG8 family
Target Tissue Specificity
Ubiquitous. Expressed at very high levels in the brain, heart, peripheral blood leukocytes, liver, kidney, placenta and skeletal muscle. Expressed at very low levels in thymus and small intestine. In the brain, expression is particularly intense in motone
Target Research Area
Neuroscience

Target Background
Enables Tat protein binding activity and ubiquitin protein ligase binding activity. Predicted to be involved in autophagosome assembly; autophagy of mitochondrion; and cellular response to nitrogen starvation. Located in autophagosome. Colocalizes with mitochondrion.
