The pipeline for the recombinant Human MAP3K14 protein production in e.coli cells consists of the following consecutive steps: construction of the expression vector encoding the Human MAP3K14 protein (400-655aa), its transformation into e.coli cells, high-density cultivation and induction of the positive cells, cell lysis, and expression analysis. The protein features a N-terminal 10xHis tag and C-terminal Myc tag fusion. The resulting recombinant Human MAP3K14 protein is purified from the cell lysate through affinity purification, and its purity is greater than 85% determined by the SDS-PAGE.

This gene encodes mitogen-activated protein kinase kinase kinase 14, which is a serine/threonine protein-kinase. This kinase binds to TRAF2 and stimulates NF-kappaB activity. It shares sequence similarity with several other MAPKK kinases. It participates in an NF-kappaB-inducing signalling cascade common to receptors of the tumour-necrosis/nerve-growth factor (TNF/NGF) family and to the interleukin-1 type-I receptor.
