Recombinant Human Keratin, type II cytoskeletal 1 (KRT1)

The DNA coding sequence translated into the human KRT1 protein sequence (2-644aa) was fused with the N-terminal 6xHis tag sequence to form the recombinant DNA, which was inserted into an expression vector. The reconstructed expression vector was transformed into the E.coli for follow-up expression. The product underwent purification to obtain the recombinant human KRT1 protein with N-terminal 6xHis tag. The SDS-PAGE analysis determined its purity higher than 85%. After electrophoresis, a 66 kDa protein band was observed on the gel.KRT1 is gene encoding a protein called Keratin, type II cytoskeletal 1 (also known as Keratin-1) in human and belongs to intermediate filament family.? Mutations in this gene have been associated with the variants of bullous congenital ichthyosiform erythroderma in which the palms and soles of the feet are affected.

The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the spinous and granular layers of the epidermis with family member KRT10 and mutations in these genes have been associated with bullous congenital ichthyosiform erythroderma. The type II cytokeratins are clustered in a region of chromosome 12q12-q13.
