CUSABIO team inserts the gene coding for the Human ODC1 protein (1-461aa) into a plasmid vector to form recombinant plasmid, which is then introduced into e.coli cells. e.coli cells demonstrating successful uptake of the recombinant plasmid are selected based on their ability to survive in the presence of a specific antibiotic. The positive e.coli cells are cultured under conditions that promote the expression of the gene of interest. A N-terminal 10xHis-SUMO tag and C-terminal Myc tag is linked to the protein. Following expression, affinity purification is used to isolate and purify the recombinant Human ODC1 protein from the cell lysate. Denaturing SDS-PAGE is then applied to resolve the resulting recombinant Human ODC1 protein, demonstrating a purity greater than 90%.

    This gene encodes the rate-limiting enzyme of the polyamine biosynthesis pathway which catalyzes ornithine to putrescine. The activity level for the enzyme varies in response to growth-promoting stimuli and exhibits a high turnover rate in comparison to other mammalian proteins. Originally localized to both chromosomes 2 and 7, the gene encoding this enzyme has been determined to be located on 2p25, with a pseudogene located on 7q31-qter. Multiple alternatively spliced transcript variants encoding distinct isoforms have been identified.
