A DNA sequence encoding the full-length human Steroid 21-hydroxylase (CYP21A2) protein was fused with a 6xHis tag at the N-terminus and then expressed in the yeast. The forming protein is the Recombinant Human CYP21A2 protein, which contains 494 amino acids. Its purity was measure by SDS-PAGE and reached up to 90%. This in-stock recombinant CYP21A2 protein is not only used for antibody synthesis but also in the studies of signal transduction. CYP21A2, a member of the microsomal cytochrome P450 family, is a critical enzyme of the glucocorticoid and mineralocorticoid biosynthesis in the adrenal cortex. Mutations in the CYP21A2 gene lead to abrupt truncations of the protein per se and further severely damage enzymatic activity, resulting in salt-wasting (SW) congenital adrenal hyperplasia (CAH).

Target Function
A cytochrome P450 monooxygenase that plays a major role in adrenal steroidogenesis. Catalyzes the hydroxylation at C-21 of progesterone and 17alpha-hydroxyprogesterone to respectively form 11-deoxycorticosterone and 11-deoxycortisol, intermediate metabolites in the biosynthetic pathway of mineralocorticoids and glucocorticoids. Mechanistically, uses molecular oxygen inserting one oxygen atom into a substrate, and reducing the second into a water molecule, with two electrons provided by NADPH via cytochrome P450 reductase (CPR; NADPH-ferrihemoprotein reductase).
