Background Information
  • Uniprot Id

    Q9NY59

  • Target Species

    Human

  • Target Name

    SMPD3

  • Target Full Name

    Sphingomyelin phosphodiesterase 3

  • Target Function

Catalyzes the hydrolysis of sphingomyelin to form ceramide and phosphocholine. Ceramide mediates numerous cellular functions, such as apoptosis and growth arrest, and is capable of regulating these 2 cellular events independently. Also hydrolyzes sphingosylphosphocholine. Regulates the cell cycle by acting as a growth suppressor in confluent cells. Probably acts as a regulator of postnatal development and participates in bone and dentin mineralization. Binds to anionic phospholipids (APLs) such as phosphatidylserine (PS) and phosphatidic acid (PA) that modulate enzymatic activity and subcellular location. May be involved in IL-1-beta-induced JNK activation in hepatocytes. May act as a mediator in transcriptional regulation of NOS2/iNOS via the NF-kappa-B activation under inflammatory conditions.

  • Target Subcellular Location

Golgi apparatus membrane; Lipid-anchor. Cell membrane; Lipid-anchor. Note=May localize to detergent-resistant subdomains of Golgi membranes of hypothalamic neurosecretory neurons (PubMed:10823942). Localizes to plasma membrane in confluent contact-inhaibited cells (PubMed:15051724).

  • Target Protein Families

    Neutral sphingomyelinase family

  • Target Tissue Specificity

    Predominantly expressed in brain.

  • Target Research Area

    Neuroscience

  • Target Synonyms

    Cca1; Confluent 3Y1 cell-associated protein 1; Neutral sphingomyelinase 2; Neutral sphingomyelinase II; NSMA2_HUMAN; nSMase-2; nSMase2; SMPD3; Sphingomyelin phosphodiesterase 3; sphingomyelin phosphodiesterase 3, neutral membrane (neutral sphingomyelinase II)

  • Target Background

Predicted to enable phosphatidic acid binding activity; phosphatidylserine binding activity; and sphingomyelin phosphodiesterase activity. Predicted to be involved in positive regulation of exosomal secretion and sphingomyelin metabolic process. Predicted to act upstream of or within several processes, including animal organ development; enzyme linked receptor protein signaling pathway; and sphingolipid metabolic process. Predicted to be located in Golgi apparatus and plasma membrane. Predicted to be active in cytoplasm. Biomarker of pulmonary emphysema.
