The pipeline for the recombinant Human DRD2 protein production in e.coli cells consists of the following consecutive steps: construction of the expression vector encoding the Human DRD2 protein (214-373aa), its transformation into e.coli cells, high-density cultivation and induction of the positive cells, cell lysis, and expression analysis. The protein features a N-terminal 10xHis tag and C-terminal Myc tag fusion. The resulting recombinant Human DRD2 protein is purified from the cell lysate through affinity purification, and its purity is greater than 85% determined by the SDS-PAGE.

Target Background
This gene encodes the D2 subtype of the dopamine receptor. This G-protein coupled receptor inhibits adenylyl cyclase activity. A missense mutation in this gene causes myoclonus dystonia; other mutations have been associated with schizophrenia. Alternative splicing of this gene results in two transcript variants encoding different isoforms. A third variant has been described, but it has not been determined whether this form is normal or due to aberrant splicing.
