The gene encoding the Human GIPR protein (22-138aa) is inserted into a plasmid vector to form the recombinant plasmid, which is introduced into baculovirus cells. baculovirus cells that have successfully taken up the recombinant plasmid can be selected based on their ability to survive in the presence of a specific antibiotic. The recombinant plasmid-containing baculovirus cells are cultured under conditions that promote the expression of the interested gene. The protein is fused with a N-terminal 10xHis tag and C-terminal Myc tag. After expression, the recombinant Human GIPR protein is isolated and purified from the cell lysate through affinity purification. The resulting recombinant Human GIPR protein is resolved by denaturing SDS-PAGE, allowing its purity to be estimated, greater than 85%.

Target Background
This gene encodes a G-protein coupled receptor for gastric inhibitory polypeptide (GIP), which was originally identified as an activity in gut extracts that inhibited gastric acid secretion and gastrin release, but subsequently was demonstrated to stimulate insulin release in the presence of elevated glucose. Mice lacking this gene exhibit higher blood glucose levels with impaired initial insulin response after oral glucose load. Defect in this gene thus may contribute to the pathogenesis of diabetes.
