Cloning the gene encoding the Human IFI6 protein (24-130aa) into a plasmid vector leads to the formation of recombinant plasmid. The resulting recombination plasmid is transformed into e.coli cells. e.coli cells containing the recombinant plasmid survive in the presence of a specific antibiotic and are selected to be cultured under conditions conducive to the expression of the gene of interest. A N-terminal 6xHis-B2M tag is linked to the protein. Following expression, the recombinant Human IFI6 protein is isolated and purified from the cell lysate using affinity purification. Denaturing SDS-PAGE is utilized to resolve the resulting recombinant Human IFI6 protein, demonstrating a purity exceeding 90%.

    This gene was first identified as one of the many genes induced by interferon. The encoded protein may play a critical role in the regulation of apoptosis. A minisatellite that consists of 26 repeats of a 12 nucleotide repeating element resembling the mammalian splice donor consensus sequence begins near the end of the second exon. Alternatively spliced transcript variants that encode different isoforms by using the two downstream repeat units as splice donor sites have been described.
